Culture-Facilitated Comparative Genomics of the Facultative Symbiont Hamiltonella defensa ArchiMer

Many insects host facultative, bacterial symbionts that confer conditional fitness benefits to their hosts. Hamiltonella defensa is a common facultative symbiont of aphids that provides protection against parasitoid wasps. Protection levels vary among strains of H. defensa that are also differentially infected by bacteriophages named APSEs. However, little is known about trait variation among strains because only one isolate has been fully sequenced. Generating complete genomes for facultative symbionts is hindered by relatively large genome sizes but low abundances in hosts like aphids that are very small. Cyanobacterial occurrence and diversity in seagrass meadows in coastal Tanzania OceanDocs

We report on the occurrence and diversity of cyanobacteria in intertidal seagrass meadows at Ocean Road and Mjimwema, Dar es Salaam, Tanzania. Nutrients, temperature and salinity were measured as comparative environmental factors. A total of 19 different cyanobacteria taxa were encountered, out of which eight were found exclusively in Mjimwema, four exclusively in Ocean Road and seven were common to both sites. Oscillatoria, Lyngbya and Spirulina were the dominant cyanobacterial genera. Cyanobacterial coverage was higher in Mjimwema (31–100%) than in Ocean Road (0–60%). The objective of this study was to evaluate the efficacy of cleaning and decontamination procedures in liquid nitrogen tanks. We evaluated 151 canisters and 133 bottoms from 133 nitrogen tanks of companies or farms for the presence of bacteria and fungi. Samples were collected from the canisters and the bottom of tanks containing liquid nitrogen. Tanks were divided into Group 1 (G1): tanks decontaminated with 2% glutaraldehyde - Glutaron® II (n = 16 canisters in 8 tanks); Group 2 (G2): decontamination with 70% ethanol (n = 20 canisters in 10 tanks); and Group 3 (G3): decontamination with 70% ethanol (n = 115 canisters in 115 tanks). Decontamination of Pig Carcasses Using Water Pressure and Lactic Acid BABT

The objective of this work was to evaluate the effect of different water pressures and concentrations of lactic acid on microbial counts (mesophilic bacteria, enterobacteria and Salmonella) on pig carcasses without contamination and contaminated carcasses, before and after the last shower and before being cooled. The tests were carried out using 4, 3 and 2 bar water pressure, and 2, 1 and 0% lactic acid concentration. In general, both the pressure in the shower and lactic acid had a positive effect by reducing the microbial count. The interaction between the pressure and lactic acid caused the largest reduction in carcasses surface count for mesophiles. Three thermophilic, anaerobic, strictly chemolithoautotrophic, sulphur- and/or thiosulphate-reducing bacteria, designated SL17(T), SL19(T) and SL22(T), were isolated from deep-sea hydrothermal samples collected at 13 degrees N (East Pacific Rise), Guaymas Basin (Gulf of California) and 23 degrees N (Mid-Atlantic Ridge), respectively. Abstract Diamondback moth (DBM), Plutella xylostella (Linnaeus), is a notorious pest of brassica crops worldwide and is resistant to all groups of insecticides. The insect system harbors diverse groups of microbiota, which in turn helps in enzymatic degradation of xenobiotic-like insecticides. The present study aimed to determine the diversity of gut microflora in DBM, quantify esterase activity and elucidate their possible role in degradation of indoxacarb. The principle of this method is to evaluate, by counts exercised upon selective media, the relative importance of the different bacteria families in the flora of the coastal waters. It is important to separate the contaminant species and the commensal species in the marine environment. Ten culture media distributed into four categories (selective, non selective, saline and non saline) were examined by numeration. Due to technical difficulties, numbers of studies dealing with benthic bacterivory are not numerous and give conflicting results. Different benthic size-compartments and their relationship to sediment chemistry in the deep Eurasian Arctic Ocean OMA

During the Arctic Expedition ARK VIII/3 (August to September 1991) with <i>RV Polarstern</i>, the macrofauna, meiofauna, Foraminifera, bacteria were sampled and sediment chemistry was determined at 13 stations along a transect from the Barents Sea slope across the deep Arctic Eurasian Basins towards the Lomonosov Ridge. Water depths ranged from 258 to 4427 m. In general, higher values for all benthic compartments as well as total organic carbon (TOC) and total hydrolysable amino acids (THAA) were recorded for the Barents Sea slope than for the deeper stations in the basins and the ridge slopes. Direct RAPD evaluation of bacteria without conventional DNA extraction BABT

The present work reports successful DNA amplification of Pantoea agglomerans and Bacillus pumilus through Random Amplified Polymorphic DNA (RAPD). For this, template DNA was obtained without conventional DNA extraction. The procedure was as follows: cultures grown for 20 hours in 5 mL LB medium were centrifuged and the resulting preparation was suspended in TE buffer. After boiling, the cell suspension was diluted and 2.0 µl were used in reactions of 15 µl. The results showed no significant differences among the RAPD profile of the PCR reactions derived from the boiling and phenol extraction methods, suggesting the utilization of this method for genetic population analysis.

Here, we describe an evaluation of fecal coliform bacteria contamination state in the ecuadorian river side area in the Provinces of Esmeraldas and Manabí, between march and may from 2008 to 2013, from an n=109 data samples. From the results, it is pretty obvious the bacteria contamination in very specific places such as Esmeraldas's river mouth, Chone's river estuary and the opposite side of the main population from the studied areas such as Esmeraldas, La Chorrera in Perdenales, Bahía de Caráquez, Jaramijó and Manta. Se evaluó el patrón de distribución espacial así como la contribución de las fracciones del plancton a la biomasa planctónica menor de 200 μm en la región NW de las aguas oceánicas adyacentes a Cuba en muestreos realizados en época de invierno (febrero de 1997 y marzo 1999). Los valores de la biomasa bacteriana variaron entre 1.6 y 11.4 mgC m-3, del nanoplancton entre 5.2 y 20.8 mgC m-3 (Xmedia = 11.4 ± 5.4 mgC m-3) y la del microzooplancton entre 4.2 y 15.5 mgC m-3 (Xmedia = 7.9 ± 3.2 mgC m-3).
